We have previously isolated and identified Pseudomonas aeruginosa N-formyl-methionyl-leucyl-phenylalanine (FMLP) as the chemotactic factor for human neutrophils. In this study, we purified an activity from the N-formyl-leucyl-methionine (FMLM)-elicited supernatant of P. aeruginosa that mediates both FMLP- and IL-8-induced chemotaxis with similar potencies. The binding activity of this activity towards FMLP and IL-8 was determined using a radioisotopic ligand-binding assay, and the potency was 3.5- and 2.2-fold higher than that of FMLP and IL-8, respectively. A comparative study with deletion mutants of N-formyl-leucyl-phenylalanine (F-Leu-Phe) designated N-formyl-leucyl-, N-acetyl-leucyl- and N-formyl-acetyl-leucyl-Phe indicated that the potency of the partial peptides was less than that of the N-formyl-methionyl-leucyl-phenylalanine (FMLP) and N-formyl-leucyl-phenylalanine (F-Leu-Phe). Furthermore, semi-quantitative analysis of competitive binding to neutrophils revealed that the competitive potency of the hydrophobic side chains of FMLP and F-Leu-Phe were similar, and that only the formyl group of F-Leu-Phe represented the critical factor for the significantly stronger potency.
